Abstract |
光第二高調波(SHG)イメージングは、これまでの手法では不可能であった生理現象の可視化能を持つにも関わらず、生体への応用が限られていた。本研究では色素の改良および応用領域の開拓の両面からSHGの生体イメージングへの展開を模索した。本研究により、既存の物とは全く異なるSHG専用色素が開発され、脳のアストロサイトと呼ばれる細胞がSHGイメージングの重要な対象となる事が明らかになった。今後の更なるSHG専用色素の開発とアストロサイトへの応用により、解析技術としてのSHGイメージングの更なる発展と、脳におけるアストロサイトの機能発現の理解が飛躍的に進むものと期待される。
Despite its unique potential to visualize biological phenomena that can hardly be achieved by other means, applications of second harmonic generation (SHG) imaging to biological researches had remained unexplored. In this study, we tried to overcome this problem by searching for novel SHG dyes as well as new biological targets to apply SHG imaging. We successfully developed new SHG dyes based on unprecedented concepts and found that astrocytes play crucial roles in the brain functions by virtue of their unique cellular characteristics, making astrocytes ideal and important new targets of SHG imaging. Further development of SHG dyes and their applications to SHG imaging of astrocytes are expected to contribute to the development of SHG imaging as well as the cellular understandings of astrocytes in the brain.
