免疫系は、病原体などを排除するための免疫応答と、自己に対する免疫応答を防ぐ免疫寛容機構の二つの仕組みにより成り立っている。免疫応答には、侵入した病原体を記憶し、次回侵入した同じ病原体を迅速に排除するためのメモリーレスポンスの仕組みが存在するが、免疫寛容機構においては記憶機構の存在は知られていない。本研究では、我々の生体内における免疫寛容機構に記憶機能が存在することを明らかにし、『memory tolerance, 免疫寛容記憶』という新しい免疫学的概念を確立することを目的として、研究をおこなっている。
申請者らはデスモグレイン3(Dsg3)特異的T細胞を用いて抗原特異的に免疫寛容機構を解析できる実験系を確立している。この系において、免疫寛容記憶は自己反応性T細胞を時間単位で排除する現象として観察される。免疫寛容記憶を担当する細胞を同定するため、記憶が成立したのちに、各種抗体を用いた細胞除去処理を行なったところ、CD3抗体処理で記憶がキャンセルされ、CD4とCD8抗体同時投与でキャンセルされなかったことから、CD3+X-Y-細胞集団が免疫寛容記憶に関連していることが推察された。さらに、フローサイトメーターの解析では、Dsg3特異的T細胞が除去される過程で、Dsg3特異的T細胞が会合する細胞集団があることが明らかとなり、同細胞がMHC class II分子陽性細胞であることがわかった。会合している細胞を蛍光顕微鏡で観察すると、MHC class II+細胞のMHC class II分子とT細胞のT細胞受容体分子が会合部位に高密度で存在している様子が観察され、抗原特異的な認識を介して会合していることが推察された。今後、CD3+X-Y-細胞集団とMHC class II+細胞の関係を明らかにし、免疫寛容記憶の責任細胞の詳細を明らかにする。
The immune system consists of two mechanisms: the immune response to eliminate pathogens and the immune tolerance mechanism to prevent the immune response against the self. The immune response has a memory response mechanism that memorizes invading pathogens and rapidly eliminates them the next time they invade, but the immune tolerance mechanism has no known memory mechanism. The aim of this study is to clarify the existence of a memory function in the immune tolerance mechanism in our body and to establish a new immunological concept of immune tolerance memory.
The applicants have established an experimental system that can analyze the immune tolerance mechanism in an antigen-specific manner using desmoglein 3 (Dsg3)-specific T cells. In this system, immune tolerance memory is observed as a phenomenon in which autoreactive T cells are eliminated on a time course. In order to identify the cells responsible for immune tolerance memory, we performed cell removal treatment using various depleting antibodies after the memory was established. This suggests that the CD3+X-Y- cell population is associated with immune tolerance memory. Furthermore, flow cytometric analysis revealed that there was a cell population of Dsg3-specific T cells that aggregated during the process of Dsg3-specific T cell removal, and these cells were found to be MHC class II molecule-positive cells. Fluorescence microscopy of the aggregating cells revealed a high density of MHC class II molecules in MHC class II+ cells and T cell receptor molecules in T cells at the aggregation site, suggesting that the aggregation is mediated by antigen-specific recognition. In the future, the relationship between the CD3+X-Y- cell population and MHC class II+ cells will be clarified to elucidate the details of the cells responsible for immune tolerance memory.
