本研究プロジェクトの目的は,薬物試験や再生移植医療のための成熟したヒト心筋組織をin vitroで構築する技術の確立である.世界に先駆けin vitroマイクロファイバー組織を開発した本塾理工学部機械工学科の尾上グループ(尾上)と,多能性幹細胞由来の再生心筋細胞の純化精製法において世界トップクラスの技術を有する本塾医学部循環器内科の福田グループ(福田,藤田,遠山)との医工連携により研究開発を実施した.
3年計画の2年目である今年は,昨年度見出した培養条件により構築したファイバ形状のヒトiPS細胞由来心筋組織に対し,電気刺激を印加することにより成熟化を試みた.結果,電気刺激デバイスを用いることで,ファイバ形状のヒトiPS由来心筋組織が同期して拍動することが確認され,組織培養を行いながら電気刺激を印加するシステムの構築に成功した.また,ハイドロゲルに内包されたファイバ状心筋組織よりmRNAを抽出し,qPCRにより遺伝子発現解析を試みた.その結果,TNNT, MYL2, Cx43などの心筋の成熟化に関わるマーカー遺伝子の発現をファイバ状心筋組織より確認した.これにより,ファイバ状iPS由来心筋組織に対する電気刺激の手法と成熟化の細胞生物学的評価法の確立を達成した.この一連の成果は国内学会において3件発表済みであり,また次年度に開催予定の国際会議microTASおよび国際学術誌に投稿準備中である.
本研究プロジェクトの最終目的である成熟したin vitro組織構築のため,最終年度となる次年度は成熟化の条件検討が主体となる.具体的には培地の栄養成分および電気刺激のパラメータを検討し,遺伝子発現・免疫染色による細胞生物学的評価の他,刺激の伝播速度や心電ポテンシャルなどの心筋組織としての指標を用いて成熟度を評価する.
The aim of this project is to establish technologies to construct in vitro matured induced pluripotent stem (iPSC)-derived human cardiac tissues for drug development and regenerative medicine. This project was launched by the collaboration between Onoe group in Mechanical Engineering and Fukuda group in Cardiology.
In this year (the second year in three-year term project), to mature fiber-shaped human iPS cell-derived cardiac muscle tissue, electrical stimulation was applied to the fiber-shaped cardiac tissue. The fiber shape human iPS-derived cardiac muscle tissue successfully contracted synchronously in response to the external electrical stimulation. In addition, the mRNA was extracted from the fiber-shaped cardiac tissues encapsulated in hydrogel, and the gene expression analysis was performed by qPCR. As a result, the expressions of marker genes involved in cardiac tissue maturation such as TNNT, MYL2 and Cx43 were confirmed from the fiber-shaped cardiac tissues. Those results indicate that we have established methods of electrical stimulation and maturation evaluation for the fiber-shaped iPS-derived cardiac muscle tissues. This series of results has been presented at three domestic conferences and is being prepared for submission to the international conference microTAS and an international journal in the next fiscal year.
In the final year (next fiscal year) of this research project, exploring optimal conditions to reconstruct a matured cardiac tissue in vitro will be the main topic. Specifically, the nutrient components of the culture medium and the parameters of electrical stimulation will be examined. In addition to the biological evaluation by gene expression and immunostaining, the propagation speed of stimulation and the electrocardiographic potential will also be analyzed for evaluating the maturation of the cardiac tissues.
