| Abstract |
ヒト大腸癌細胞株WiDr(親株)に5-fluorouracil(5-FU)を曝露することで, 耐性が出現した細胞株(耐性株)を樹立した。耐性株では, 親株と比較してHeat Shock Protein 27(HSP27)の発現が増強していた。耐性株において増強したHSP27を再抑制することで, 一旦出現した耐性の緩和を認めた。特に大腸癌化学療法中の耐性出現時における, HSP27を標的とした新たな治療の開発に向けた重要なデータである。
We generated the drug-resistant subclone of human colon cancer cell line WiDr by exposure to 5-fluorouracil (5-FU). Higher Heat Shock Protein 27 (HSP27) level was demonstrated in drug-resistant subclone than in naïve cell line. Down-regulation of overexpressed HSP27 suppressed generated 5-FU resistance in drug-resistant subclone. These data is important to develop a new therapy targeted on HSP27, especially to overcome generated resistance during chemotherapy in colon cancer.
