Abstract |
体細胞を起点として幹細胞を経ずに目的の細胞を直接的に誘導する手法(直接リプログラミング法)が, 幹細胞を用いない新たな再生医療の手法として近年報告されている。我々はマウス線維芽細胞に3または4個の特異的因子遺伝子を強制発現させ培養した細胞が, II型肺胞上皮細胞に特異的なマーカーであるSurfactant Protein C(SP-C)を発現していることを免疫染色で確認した。さらにそれらの細胞内にII型肺胞上皮細胞に特異的とされるLamellar bodyの存在を電子顕微鏡で確認した。これらの結果から, 直接リプログラミング法によってII型肺胞上皮細胞が誘導されたことが示唆された。
Direct reprogramming has been reported recently as one of the new methods of regenerative medicine. By direct reprogramming method, somatic cells are converted into other types of cells directly, not using stem cells. We induced 3 or 4 selected factors into fibroblasts of mice. After culturing, the induced cells were found to express Surfactant Protein C, which is a specific marker of type II alveolar cell (AT II) by immunofluorescence staining. The presence of "lamellar body" , which is known as a specific structure of AT II, was detected in the cytoplasm of these induced cells by electron microscopy. These results suggested that induced AT II was obtained by direct reprogramming method.
