| Abstract |
心室形成不全の表現型を持つメダカ変異体動物を化学変異剤により作製、ポジショナルクローニング法により原因遺伝子の同定と作用機序の解明を行った。
ポジショナルクローニング法により原因遺伝子は、versicanというコンドロイチン硫酸プロテオグリカンであるという結果を得た。プロテオグリカンは脊椎動物の細胞外マトリックスで生物活性など持たない物理的な支えと考えられてきたが、カエデ蛍光蛋白の導入実験成果から、接触している心筋幹細胞の移動を能動的に制御して微小環境を形成していることが分かった。心室形成のためには心筒形成後に外部からの心臓幹細胞の流入が必要であることを発見した。
