| Abstract |
本研究の目的は, クロマチン構造を変化させる初期化因子の一つと考えられる卵子特異的リンカーヒストンH1fooに注目して, その機能を明らかにすることにより, 哺乳類卵子におけるクロマチンのエピゲノム構造の解明をはかった。卵子幹細胞と思われる細胞系には卵子特異的リンカーヒストンの発現を検討したもののその発現は認められなかった。さらに卵子への分化におけるヌクレオソーム構造の変化についての役割を検討した。卵子特異的リンカーヒストンは, 卵子への分化に重要な役割を有している可能性が示唆された。
Oocyte-specific linker histone H1foo is expressed in oocyte and early stage embryos. H1foo is essential for in vitro meiotic maturation of oocyte. The greater mobility of H1foo, compared with H1, may contribute to this rapid replacement and the instability of chromatin structure. These findings suggest that the rapid replacement of H1 with H1foo may play an important role in nuclear remodeling.
