Abstract |
糖尿病発症db/dbマウスの27%で胃排出が延長した。胃排出遅延db/dbマウスで胃筋間神経叢のnNOSとGFAP発現が低下した。胃排出遅延db/dbマウスで血清ApoE値が有意に低下した。ApoE KOマウスでは胃排出が低下し、胃排出遅延db/dbマウスと同様、筋間神経叢のnNOSやGFAP発現が低下した。nNOS KOマウス、ApoE KOマウスに神経幹細胞移植6週後の胃排出能は移植前に比べ有意に改善した。ApoE KOマウスでnNOSとGFAP発現は有意に回復した。本研究は神経幹細胞移植が胃運動不全麻痺で胃排出遅延を是正し、血清ApoEが消化管運動障害のマーカーとなることが示唆された。
Delayed gastric emptying was observed in 27 % of db/db mouse with a significant decrease in the serum ApoE level and in the myenteric nNOS and GFAP expression. The gastric emptying was also significantly delayed in ApoE knockout (KO) mouse with a decrease in the expression of nNOS and GFAP in myenteric plexus. Transplantation of neural stem cells (NSCs) improved delayed gastric emptying in nNOS KO mice and ApoE KO mise. The expressions of nNOS and GFAP in myenteric plexus were restored by transplantation of NSCs. Transplantation of NSCs restored the delayed gastric emptying in apoE knockout mice, suggesting a promising application of regenerative therapy against gastroparesis and a possible development of serum ApoE as a potential biomarker for gastrointestinal motility disorders.
