| Abstract |
進行・再発非小細胞肺がん患者の標準治療の1つである「ドセタキセル・カルボプラチン併用療法」について、高齢者に対するドセタキセルの至適投与量・用量調節指標の探索を行った。その結果、年齢の上昇に伴ってドセタキセルのクリアランスが低下する(曝露量が増加する)傾向が認められた。しかしながらクリアランスの個体間バラツキが大きいため、一概に高齢者での減量を推奨することはできない。またドセタキセルの効果および副作用は遊離形濃度に依存するため、血中α1酸性糖蛋白質(α1-acid glycoprotein;AGP)濃度に影響を受ける。以上より、ドセタキセルの高齢者への至適投与量を考慮する際の用量調節指標として、年齢のみならず血中AGP濃度も含む複数因子の妥当性が示唆された。
