シロアリなどの真社会性昆虫では、個体は発生過程において繁殖カースト(繁殖を行う個体)か非繁殖カースト(繁殖以外の仕事を行う個体)のいずれかに分化し、集団内の個体間で繁殖に関する分業がみられる。本研究ではヤマトシロアリを材料とし、繁殖・非繁殖カーストの分化決定に遺伝子が関わるか解明するため、それらの間での遺伝的変異の検出を行うことを目的とした。本種の繁殖・非繁殖カーストを合計で108個体用い、それらのDNAを使ってゲノムワイドな遺伝子解析技術であるRestriction site Associated DNA Sequencing (RAD-seq) を行い、カースト間の遺伝的変異の調査をした。RAD-seqデータのコンピューター解析の結果、42,663のゲノム領域においていずれかの個体間で遺伝的変異があることを確認した。それらの遺伝的変異のうち、16領域においては繁殖・非繁殖カースト間で明確な違いがあった。この16領域についてカースト間の遺伝的変異を追認するために、これらの領域にPCRプライマーを設計し、カースト間でPCR増幅に違いが生じるか調べた。これらの領域のうち、1領域において(遺伝子領域コード:RADtag463764)はカースト間で明白なPCR増幅の違いがみられた。この結果は、繁殖・非繁殖カースト間においてゲノムに差異があることを示しており、その領域またはその近傍領域の遺伝子がカースト分化決定に影響を与えていることを示唆する。今後は、これらのゲノム領域に存在する遺伝子の塩基配列決定や機能解析を行うことで、カースト分化決定に関わる遺伝子を同定することができる。繁殖・非繁殖カーストの分化とそれらの分業は真社会性の根幹を成すものであり、本研究をさらに発展させることで、真社会性の進化過程や社会組織化機構の解明につながる。
In eusocial insects such as termites, individuals differentiate into either reproductive caste (individuals that reproduce) or non-reproductive caste (individuals that perform tasks other than reproduction) during development, and a division of labor regarding reproduction is established among individuals in a social group. In this study, using the Japanese termite Reticulitermes speratus, I aimed to detect genetic variation among them in order to elucidate whether genes are involved in caste determination and differentiation between reproductive and nonreproductive castes. A total of 108 reproductive and non-reproductive castes of this species were used to investigate genetic variation among the castes using a genome-wide analysis technique, Restriction site Associated DNA Sequencing (RAD-seq). A computer analysis of the data confirm the presence of genetic variation between any of the individuals in 42,663 genomic regions. Of these genetic regions, 16 showed clear differences between reproductive and non-reproductive castes. To reconfirm the genetic variation between the castes in these 16 regions, I designed PCR primers for these regions and examined if caste-specific PCR amplification occurred. In one of these regions (genomic region code: RADtag463764), there was a clear difference in PCR amplification between the castes. This result indicates that there are genomic differences between reproductive and non-reproductive castes, and suggests that genes in or near this region influence caste determination. Future sequencing and functional analysis of the genes in these genomic regions will help to identify the genes involved in the caste determination. The differentiation of reproductive and non-reproductive castes and the division of labor between them form the basis of eusociality, and further studies will contribute to the elucidation of the evolutionary process and social organization mechanism of eusociality.
