食道癌においてfibrinogenは予後不良因子の1つである. Fibrinogenの受容体であるインテグリンβ3 (CD61)陽性細胞は, EGF, bFGF含有無血清培地による培養において, sphere様細胞塊の形成により自己複製能が示された. CD61陽性細胞において, 走化能が高い傾向が示唆された. 一方, 食道扁平上皮癌細胞株であるTE10およびTE11を用いて抗癌剤(5FU)への長期曝露による癌幹細胞の純化を行った. miRNA Arrayを行った結果, 幹細胞活性に関係するmiRNA (miR-200ファミリーもしくはmiR-let7ファミリー)の発現変動が検出された.
Fibrinogen is one of the prognostic factors in esophageal cancer. In esophageal squamous carcinoma cell lines, integrin beta 3 (CD61/fibrinogen receptor) positive cell line (TE8) showed sphere-formation in serum-free medium containing growth factors (EGF and bFGF). This result suggested that CD61- positive cells have the property of self-renewal of cancer stem cell. Furthermore, the analysis of cell morphology functions, CD61-positive cells indicated the high chemotactic ability. On the other hand, to enrich for cancer stem cells, followed by purification of cancer stem cells by long-term exposure to anti-cancer agents (5FU) in esophageal squamous carcinoma cell lines TE10 and TE11. After six months, both cells have acquired the ability for drug resistance, and the expression variation of miRNAs (miR-200 family or miR-let-7 family) were detected.
