細胞内のオルガネラのpHは、それぞれのオルガネラで厳密にコントロールされており、種々の生化学反応が制御されている。オルガネラの一つであるリソソームは、エンドサイトーシスによって取り込んだものや細胞成分を分解利用するのが役割である。リソソーム内のpHは5程度と弱酸性であり、その内部には酸性で活性化する様々な加水分解酵素が存在し生体成分を分解している。また、リソソーム病においては、リソソーム内pHの異常が疾病に繋がるとの報告もある。そのため、リソソーム内のpHを測定することは、生体メカニズムの解明のみならず、疾病メカニズムの解明、さらにはそれらの治療薬の開発へと繋がることが期待される。本研究では、生きた細胞内で、リアルタイムにリソソーム内pHの定量を可能とする新たな蛍光プローブの開発を目的とした。
これまでに開発しているレシオ型pH蛍光プローブであるSiRpH類(J. Am. Chem. Soc., 140, 5925 (2018))を基礎骨格として、その分子構造を改良することで、リソソーム内pHの測定に適した低いpKaを持ち、汎用される蛍光顕微鏡のレーザー波長に適した新たなレシオ型pH蛍光プローブの開発に成功した。さらに本pH蛍光プローブをデキストランに標識し、それをリソソームのマーカータンパク質であるTmem192(EGFP融合)を安定発現させたMEF細胞に導入し、生細胞イメージングを行った結果、マーカータンパク質とプローブの蛍光は高い重なりを示し、リソソーム内の平均pHは5.1程度であり、これまでの報告と矛盾のない値であった。さらに、本pH蛍光プローブの生命科学研究における有用性を示すために、細胞生物学の専門家である、東北大学大学院生命科学研究科・田口友彦 教授との共同研究によって、開発したデキストランに標識したpH蛍光プローブの有用性を評価しており、光褪色耐性の高さや計測したpH値の精度の高さなどの既存の蛍光プローブに対する有用性が見いだされている。
The organelle pH inside cells is strictly controlled in each organelle, regulating various biochemical reactions. Lysosome, which is one of organelles, play a role for the degradation and the recyclization of substances taken in by endocytosis and cellular components. The pH inside lysosomes is reported to be around 5, which is weakly acidic, and there are various hydrolases, which can be activated in acidic conditions, degradating biological components. Moreover, there are some reports that the abnormal pH in lysosomes leads to the human diseases as the lysosomal diseases. So, the measurement of lysosomal pH is expected to lead to the elucidation of biological and pathological mechanisms, and the development of drugs for these diseases. In this study, we aimed to develop a novel fluorescence probe for quantifying lysosomal pH in living cells in real time.
So far, we successfully developed a novel ratiometric fluorescence probe, which has a suitable pKa for measuring the acidic pH and an absorption wavelength suitable for the light wavelengths of widely used lasers equipped with the fluorescence microscopes, based on our previously developed ratiometric pH fluorescence probes, SiRpHs (J. Am. Chem. Soc., 140, 5925 (2018)) as a basic scaffold. Further, this developed pH fluorescence probe was labelled to dextran. This dextran conjugates were introduced to MEF cells expressing lysosomal marker protein (Tmem192) fused with GFP, and the live-cell fluorescence imaging was performed. As a result, the fluorescence of the fluorescence probe was well merged with that of lysosome marker proteins, and the average lysosomal pH was measured to be around 5.1, which is consistent with the previous reports. Furthermore, to demonstrate the usefulness of this fluorescence probe for life science research, the functions of the dextran conjugates labelled with the developed pH fluorescence probes is now being examined through the collaboration with Prof. Tomohiko Taguchi (Graduate School of Life Science, Tohoku University) as an expert for the cell biology. So far, high photostability, the accuracy of measured pH values, etc. of the developed fluorescence probe are observed as advantages to the existing pH probes.
