我々は前立腺がん幹細胞に発現しているCXorf48を標的としたがん免疫療法の開発を目指している。そのためにCXorf48特異的な診断用モノクローナル抗体の開発と、CXorf48を標的とする遺伝子改変T細胞の作製を行った。
まず、精製したMBP-CXorf48タンパク質とアジュバントをBALB/cCrSlcマウスに免疫し、後腸骨リンパ節由来のリンパ球とミエローマ細胞(SP2)からハイブリドーマを作製した。CXorf48発現HEK293T細胞を用いたIn Cell Western法によりハイブリドーマの培養上清の抗原反応性を評価し61種の細胞を選出した。さらに上位7クローンについて限界希釈法によるクローニングを行い4種の有望なクローンを得た。今後、選別したモノクローナル抗体を用いて、前立腺がん細胞株より EOS システムを用いて分離した幹細胞における CXorf48抗原の発現を検討する。
また、我々はこれまでにHLA-A*24:02 拘束性に提示されるエピトープを認識するT細胞受容体(TCR)遺伝子導入T細胞を作成しているが、予想していたよりも抗原認識能が低かった。これは内在性TCRと導入されたTCRのミスペアリングにより抗原認識能が低下していた可能性があるため、本研究では、内在性TCRの発現を低下させるsiTCR発現レトロウイルスベクターを用いて再度TCR-T細胞を作製した。すなわち、CXorf48特異的TCRα鎖およびβ鎖遺伝子のVJ領域およびVDJ領域をsiTCRベクターに組み込んでヒトリンパ球に導入し、TCR-T細胞の抗原認識能を評価した。CXorf48,siTCR導入ヒトリンパ球はCXorf48ペプチドを添加したC1R-A24細胞に対して高いIFN-γ産生能を示した。また、CXorf48,siTCR導入細胞は、CXorf48とHLA-A*24:02を高発現しているK562細胞(K562-A24)に対しても高いIFN-γの産生を認めた。さらに、51Cr細胞傷害性試験により、K562-A24細胞への傷害能も確認された。今後は本遺伝子改変T細胞を用いて前立腺がん幹細胞に対する傷害性を明らかにしていく。
We are aiming at development of strong immunotherapy against CXorf48, which is highly expressed in prostate cancer stem cells. For this purpose, we have made monoclonal antibody specific to CXorf48 and also generated the gene-modified T cells against this antigen.
First, we immunized mice with MBP-CXorf48 protein and generated hybridoma using lymphocytes and myeloma cells. We screened these hybridomas by in cell western method using CXorf48 gene-transduced HEK293T cells, and finally obtained 4 clones. We are going to stain prostate stem cells with these antibodies.
On the other hand, we have previously made anti-CXorf48 T cell receptor(TCR)-T cells which recognize HLA-A*24:02-restricted epitope, however, their antigen recognition was relatively weak due to mispairing of internal TCR and transduced-TCR. In this study, we made new retroviral vector which contain siTCR and anti-CXorf48 TCR. Human T cells transduced with this vector recognized CIR-A24 cells pulsed with CXorf48 peptides, and also showed cytotoxicity against K562 cells which have both HLA-A*24:02 and CXorf48. We are going to evaluate cytotoxicity of the TCR-T cells against prostate cancer stem cells.
