Abstract |
本研究では、骨疾患治療用バイオマテリアルの開発を目的とし、生体膜由来のベシクルであるリポソーム表面に多糖、DNAなどの生体由来高分子を交互に積層化し、さらにその表面へリン酸カルシウム(CaP)を析出することによって、有機・無機ハイブリッドリポナノカプセルを作製した。この際、反応条件を変化することにより、CaP結晶構造(結晶種、厚みなど)の調節を行い、ナノカプセルからの物質の放出とナノカプセルの骨組織内への取り込みについて制御を試みた。さらに、CaP層表面にDNAを骨標的リガンドとして提示させたところ、骨のモデルであるハイドロキシアパタイトに対して高い集積性を示すようになった。
As an approach to create a bone therapeutic biomaterial, we created an organic-inorganic hybrid nanocapsule by utilizing polysaccharide-coated liposomes as a reaction site for the deposition of calcium phosphate (CaP). By tuning the reaction conditions and the surface chemical composition of nanocapsules, it was possible to control in mineralization, such as thickness and crystal structures, over the nanocapsules. Furthermore, DNA was selected as a bone-targeting moiety and it was adsorbed over the nanocapsules. DNA-coated nanocapsules exhibited a selective accumulation onto the powdery hydroxyapatite, which is the main constituent of bone and teeth.
