Abstract 
正常腸管上皮幹細胞マーカー遺伝子であるLGR5を標的とし、蛍光蛋白レポーターおよびlineage tracerであるCreERのヒト腸管上皮オルガノイドへのKnock-inを試みた。
最新のゲノム編集技術であるCRISPR/Cas9のヒト腸管上皮オルガノイドへの導入の最適化を行い、オルガノイドにおける遺伝子相同組換え技術を確立した。その技術を利用し、LGR5遺伝子座へのCreER のKnock-inに成功した。 
To elucidate the role of Lgr5 stem cells in colorectal cancer, we attempted to knock-in fluorescence reporter and CreER (liniage tracer) in an Lgr5 locus. Taking advantage of genome editing system, CRISPR/Cas9, we found an optimal condition to engineer human intestinal organoids. With this system, we succeeded in knock-in of CreER into the Lgr5 locus.
