Abstract |
炎症性腸疾患は慢性腸管炎症を特徴とする疾患で, 本邦で近年増加中である。潰瘍性大腸炎患者は長期罹患とともに大腸発がんリスクが高くなり,通常の大腸がんと異なる臨床像を示し, Colitis associated cancer(CAC)と称される。我々はCAC症例より腫瘍サンプルを採取し, オルガノイドの樹立,そのCACオルガノイドの統合的な解析を行った。CACでは通常型の大腸がんに比し, 低頻度のAPC変異と高頻度のTP53変異を認めた。CACオルガノイドの樹立とその分子遺伝学的解析データが統合され, 今後のCACに対する基礎研究ならびにトランスレーションナルリサーチの進展に極めて有用と考えられる。
Inflammatory bowel disease is characterized by chronic mucosal inflammation and the number of patients are increasing in our country. Duration of disease is positively correlated to the occurrence of colorectal cancer, which is referred to as Colitis associated cancer (CAC). CAC exhibit different clinical features and therapeutic response in comparison to sporadic colorectal cancer. To elucidate carcinogenic mechanism for CAC, we attempted to establish organoids from CAC. We established 6 CAC organoids and performed gene expression microarray analysis, exome analysis and copy number variation analysis. We observed less frequent mutations in APC and higher prevalence of TP53 mutations in CAC as compared to sporadic colorectal cancer. The established CAC organoids and their molecular characterization data can be useful resource for basic and translational research for CAC.
