Abstract |
腸管内でのTregの動態や機能制御を詳細に1細胞レベルで解析した研究はない。また、潰瘍性大腸炎患者では粘膜内Tregは健常腸管と比してTregが増加していることが報告されている。一方で、本患者で腸管炎症の慢性化・上皮修復不全を示す。これは潰瘍性大腸炎ではTregの分化・機能異常ではなく、局在異常により、上皮修復を効率的に行えていない可能性を示唆している。Tregの消化管疾患時における動態を解析するため、本研究では IVMを用い、in vivoモデルでの炎症時におけるTregの腸管粘膜内局在(粘膜固有層or腸管上皮間)、粘膜固有層-上皮間の相互移行の頻度や移行の動態を検討した。
The dynamics and functional regulation of Treg in the intestine at the single cell level is unknown. It has been reported that the number of Treg in the mucosa is increased in patients with ulcerative colitis compared with that in healthy intestine. On the other hand, chronicity of intestinal inflammation and insufficient epithelial repair are shown in this patient. In order to analyze the dynamics of Treg in gastrointestinal diseases, this study used IVM to investigate the localization of Treg in the intestinal mucosa during inflammation in an in vivo model. In this study, we used IVM to examine the frequency and dynamics of intramucosal localization of Treg (between the mucosal lamina propria and intestinal epithelium) and mucosal lamina propria-epithelium cross-migration during inflammation in an in vivo model.
