Abstract |
EZH2の選択的メチル化抑制阻害剤である GSK126を用いて直接的な抗腫瘍効果の検討を行った。ウエスタンブロット法にて, ヒト膀胱癌細胞株(UMUC3, KU19-19, T24, 5637)におけるEZH2の発現は確認できたが, GSK126投与後のH3K27me3発現の変化は認められなかった。さらにWSTアッセイでは濃度・時間依存ともにGSK126の有効な殺細胞効果は認められなかった。EZH2抑制と免疫チェックポイント阻害との相乗効果が検討されているが, 上部尿路上皮癌検体においてPD-1発現を検討したところ, 腫瘍内部のPD-1強陽性が独立して上部尿路上皮癌死と関連していた。
We examined direct antitumor effect for urothelial carcinoma by using GSK126, which is an inhibitor of selective methylation of EZH2. Expression of EZH2 could be confirmed in human bladder cancer cell lines (UMUC3, KU19-19, T24, 5637) by the Western blotting method, whereas expression of H3K27me3 after administration of GSK126 in vitro was no change in the expression level compared with the control. Furthermore, we examined the effect of GSK on cell growth by WST assay, and no effective decrease of cell viability in either concentration dependence or time dependence. Recently, a synergistic effect between inhibition of EZH2 and immunocheckpoint inhibitor has been studied.We examined the role of PD-1 expression on the clinical outcome of upper tract urothelial carcinoma. PD-1 positivity in the tumor inside could be a strong predictor for a worse clinical outcome.
