Abstract |
酸素透過性のポリジメチルシロキサン(poly-dimethylsiloxane ; PDMS)膜を底面 とする特殊な培養器(PDMS培養器)を用いることによって, 心筋塊(スフェロイド)の作製が可能となった。これらの心筋細胞は, 成熟化マーカーであるmyosin light chain (MLY2)が高い傾向にあり, 心筋細胞が成熟化傾向にあった。一方, 酸素透過しない従来の培養器では, 低酸素誘導因子(HIF : Hypoxia-Inducible Factor)のtarget遺伝子発現が高い傾向が確認された。
By using a special incubator with an oxygen permeable poly-dimethylsiloxane (PDMS) membrane as the bottom surface, it was possible to product myocardial spheroids. These cardiomyocytes tended to have a high myosin light chain (MLY 2) which is a maturation marker. On the other hand, in the conventional incubator that does not permeate oxygen, the tendency of target gene expression of Hypoxia-Inducible Factor (HIF) was high.
