閉経によるエストロゲン欠乏は, 骨を吸収する破骨細胞の活性化から骨量減少をきたし, 骨粗鬆症を発症する。申請者は, 低酸素応答性の代表的な転写因子であるHIF1αが, エストロゲン欠乏により破骨細胞に発現することを見出した。そこで, 閉経後骨粗鬆症治療薬の候補として, 破骨細胞のHIF1αを抑制する活性がある薬剤を同定した(2016年論文報告)。また, 男性骨粗鬆症モデルにおいても, 破骨細胞にHIF1αが蓄積することを見出し(2016年論文報告), 同定した破骨細胞のHIF1αの阻害剤のいずれもが, 男性骨粗鬆症モデルによる骨量減少を完全に抑制できることを明らかにした(2017年論文報告)。
Estrogen deficiency due to menopause causes activation of osteoclastic bone-resorption leading to osteoporosis development. We found that HIF1α, a hypoxia responsive transcription factor, was expressed in osteoclasts under an estrogen deficient condition. Then, we screened and identified several HIF1α inhibitors as candidate therapeutic agents for postmenopausal osteoporosis treatment (published in 2016). We also found that HIF1α was expressed in osteoclasts in male osteoporosis model mice (published in 2016). We demonstrated that reduction of bone mass in male osteoporosis model mice could be completely abrogated by administration of the HIF1α inhibiting agents, all of which were identified by us (published in 2017).
