本研究では、生体膜由来のカプセル素材であるリポソームの表層をデザインすることで、分子の特性(分子量、親疎水性、荷電性)に合わせた薬剤の保持・放出能の付与と皮膚浸透性の発現を狙い、経皮薬物送達システム(Transdermal drug delivery system, TDDS)の開発を目指している。本年度は、リン脂質とともにラフト形成能を有するグルコシルセラミド(GlcCer)を用いてリポソームを作製し、その表面にバイオポリマーの積層化を行うことで種々の水溶性物質と油溶性物質の封入を行い、環境変化に応じた放出の制御を試みた。
まず、GlcCerとリン脂質を用いて、粒径約80nmのリポソームを作製した(GlcCerLipo[+])。次に、カチオン性多糖であるキトサンをリン酸化することで両性のリン酸化キトサン(PCHI)を合成し、GlcCerLipo[+]の表面に積層化した(GlcCerLipo[+]-PCHI)。リポソーム(Lipo[+])と比較してGlcCerLipo[+]とGlcCerLipo[+]-PCHIでは、膜強度が向上することがわかった。また、GlcCerLipo[+]-PCHIは、pHに応じてゼータ電位が変化し、アミノ基とリン酸基に由来する両性を示した。pHによってカプセルの荷電状態を調節することで、ポリマー層にアニオン性とカチオン性の物質をそれぞれ封入することができた。さらに、pHを変化させることで、放出を調節することができた。また、リポソーム表面にバイオポリマーとその分解酵素を積層化することで、ポリマーの酵素分解に伴って物質を放出させることができた。
次に、DMSOと水の混合溶液にGlcCerLipo[+]-PCHIを分散させたところ、Lipo[+]は低濃度のDMSO水溶液中で崩壊した。一方、GlcCerLipo[+]-PCHIは、90vol%DMSO水溶液中でも粒径を維持し、耐溶剤性の向上が見られた。そこで、DMSOに溶かした油溶性色素を添加したところ、カプセルは崩壊せず、高い保持性を示した。この際、溶液中に少量の界面活性剤を添加して疎水場を形成させることで、色素を放出させることができた。
以上のように、対象物質に対するポリマー層の親和性を調節することで、物質の封入と保持、さらに環境条件によって親和性を調節することで放出を制御することができた。
In this study, we prepared biobased nanocapsules by the polymer deposition over the ceramide-containing liposomes to render abilities to load and release versatile substances to aim for transdermal drug delivery.
First, we prepared a liposome composed of glucosylceramide (GlcCer) and phospholipids having a hydrodynamic diameter of approximately 80 nm. Next, phosphorylated chitosan (PCHI) was synthesized by reaction of chitosan (CHI), with phosphorus pentoxide. By depositing PCHI onto GlcCerLipo[+], an amphoteric capsule (GlcCerLipo[+]-PCHI) was obtained. The structural stability of GlcCerLipo[+] and GlcCerLipo[+]-PCHI was found to be improved compared to Lipo[+]. Anionic and cationic substances could be incorporated by adjusting the charge of GlcCerLipo[+]-PCHI to be positive and negative, respectively. Their release was tuned by controlling the pH value. Next, lysozyme, which is an enzyme that can degrade CHI, was deposited onto GlcCerLipo[+]-PCHI to obtain a capsule (GlcCerLipo[+]-PCHI-Lyso) with the enzymatic degradability. It was found that the number of fragments of degraded PCHI was significantly increased at the optimal condition for Lyso, and that the release of cargos was triggered by the enzymatic degradation. Next, we evaluated the structural stability of GlcCerLipo[+]-PCHI in the mixture of water and DMSO by measuring the change in turbidity and Dh of capsules. It is noteworthy that GlcCerLipo[+]-PCHI maintained its size even in 90vol% DMSO solution whereas Lipo[+] showed a significantly low solvent resistance. These results suggest that the inclusion of ceramide in the lipid membrane and the formation of PCHI layer could allow for the improved structural stability and high organic solvent resistance. In fact, we could load a lipophilic dye into GlcCerLipo[+]-PCHI in the DMSO aqueous solution (50vol%) without rupturing. Next, the dye could be released from GlcCerLipo[+]-PCHI by adding a small amount of a surfactant. We expect that the obtained liposome-based nanocapsules will be applied for a transdermal delivery of various drugs/actives.
