本研究では、場所によって異なる挙動を示す肝臓を対象に、薬剤によって肝臓が慢性化していく動的な変化を再現するシミュレーションモデルの開発を行った。既存のSPARKライブラリを用いた解析では細胞の挙動の再現に限界があるために、Python言語+Mesaライブラリを用いて、細胞の動きをエージェントとして実装しなおした。従来のモデルと比較し、門脈から薬剤が注入され中心静脈で細胞死が起きる現象の再現、細胞死の時点で周辺に分泌する物質の再現、中心静脈からの位置によって肝細胞の薬剤感受性を変更するなどを実装できた。特に肝臓の形状をどのような形にでもできる仕組み、感受性と位置に関して様々な関数など自由度の高い設計としたため、今後様々な仮説の設立などを行いやすい仕組みとした。また、これらの評価データを得るために、従来の培養ではなく、肝臓の場所ごとに起きている酸素濃度の勾配を再現している特殊な培養系を用いて、それぞれの細胞を薬剤に暴露し、その後の代謝物の濃度変化を調べた。キャピラリー電気泳動・質量分析装置を用いてメタボローム解析によって網羅的なイオン性代謝物を取得し、論文を出版した。これらは東京大学酒井博士らとの共同研究の成果である。
In this study, we developed a simulation model for the liver, which behaves differently in different locations, to reproduce the dynamic changes that occur as the liver becomes chronic due to drugs. Since the existing SPARK library analysis has limitations in reproducing cell behavior, we re-implemented the cell motion as an agent using the Python language + Mesa library. Compared to the existing model, we were able to reproduce the phenomenon in which cell death occurs in the central vein after drug injection from the portal vein, reproduce substances secreted in the surrounding area at the time of cell death, and change the drug sensitivity of hepatocytes according to their position from the central vein. In particular, we designed a mechanism that allows the liver to be shaped in various ways and a variety of functions for sensitivity and position, which facilitates the establishment of various hypotheses in the future. To obtain these evaluation data, we exposed each cell to the drug using an original culture system that reproduced the gradient of oxygen concentration occurring in each location of the liver, rather than a conventional culture and examined the subsequent changes in metabolite concentrations. Comprehensive ionic metabolites were obtained by metabolomic analysis using capillary electrophoresis and mass spectrometry, and a paper was published. These are the results of collaborative research with Dr. Sakai and colleagues at the University of Tokyo.
