ウイルスの中でもバクテリアに特異的に感染するウイルスのことをファージと言い、その形態はいわゆるウイルス(新型コロナウイルスやインフルエンザウイルス等)とは大きく異なり、遺伝物質が格納される頭部と、感染に寄与する尾部から構成される。近年、様々なバクテリアにおいて、頭部を欠き(つまり増殖能力はない)、尾部のみからなるファージ様タンパク質の生産が報告されている。しかしながら、多くのバクテリアがこのようにファージ様タンパク質を生産する生理学的の意義の全貌は未だわかっておらず、本研究では、その詳細な解析を行い、将来的にはドラッグデリバリーシステムへ応用することを目的としている。
本研究に用いたStreptomyces属放線菌のゲノムを相同性検索したところ、ファージ様タンパク質をコードする遺伝子群が一連のクラスターを形成していることが示唆された。しかしながら、これらを構成する各遺伝子の正確な役割は未知であったため、ターゲットタンパク質をコードする遺伝子クラスター中の遺伝子の一部を破壊し、ファージ様タンパク質の生産機構や、生理学的意義を検討することとした。
遺伝子破壊はゲノム編集(CRISPER-Cas9)により行った。CRISPER-Cas9で切断できる領域は制限があるため、候補を4つに絞りゲノム編集を試みた。
まず、大腸菌を用いてゲノム編集に用いる4種類のベクターを構築した。このベクターを接合伝達によりStreptomyces属放線菌に導入し、Cas9を誘導するための抗生物質を添加した培地にて複数回植え継いだ。その後、コロニーPCRにより編集状況を確認した。しかしながら、ゲノムが編集された株は取得できなかった。この原因として、編集すると致死になる、Cas9を誘導するプロモーターの機能が弱い、等が考えられた。したがって現在、Cas9の誘導プロモーターを改変し、再度ゲノム編集を行なっている。また、編集位置の変更も試みている。
Viruses that specifically infect bacteria are called phage, and their morphology differs significantly from that of so-called viruses (e.g., novel coronaviruses and influenza viruses) in that they are composed of a head, in which genetic material is stored, and a tail which contributes to infection. In recent years, the production of phage tail-like proteins that lack a head (i.e., have no ability to multiply) and consist only of a tail has been reported in a variety of bacteria. However, the physiological significance of this phage tail-like protein production in many bacteria is not yet fully understood, and this study aims to analyze it in detail and apply it to drug delivery systems in the future.
A homology search of the genome of Streptomyces species used in this study suggested that a group of genes encoding phage tail-like proteins are composed of clusters. However, since the exact physiological role of each gene in these clusters was unknown, we decided to disrupt some of the genes in the gene clusters encoding the target proteins to investigate the production mechanism and physiological significance of the phage tail-like proteins.
The gene disruption was performed by genome editing (CRISPER-Cas9), and since the regions that can be disrupted by CRISPER-Cas9 are limited, we narrowed down the candidates to four and attempted to perform genome editing.
First, four types of vectors for genome editing were constructed using E. coli. These vectors were introduced into Streptomyces species by cojugation, and inoculated several times in a medium with antibiotics to induce Cas9. The editing status was then confirmed by colony PCR. However, no genome-edited strains could be obtained. The reasons for this were thought to be that editing was lethal, the promoter function to induce Cas9 was weak, and so on. Therefore, the Cas9 inducing promoter is currently being modified and the genome is being edited again. We are also trying to change the position of genome editing.
