Optical coherence tomography (光CT)は半透明物質の深さ方向の画像を非侵襲で計測する手法であり、臨床応用されている。光CTでは玉ねぎ状の多層物体を構成する界面の反射率を計測する機器である。光CTの基本的な原理は、広帯域光の可干渉距離が短い事を用い、深さ方向の反射率分布を干渉計測で取得し、横方向には光ビームを照射する場所をスキャンすることで断層像計測をおこなう。言うまでもないことであるが、界面の反射率以上の情報が得られる方が様々な応用を行う上で好ましい。私たちは、広帯域光を用いた干渉計測を用いて被測定物内部の界面における揺らぎスペクトルを計測する手法の開発を行った。すべての物体内部の原子や分子は熱エネルギーにより乱雑な運動を行っている。この熱運動のスペクトルを計測すると被測定物の粘度や表面張力などの物性を測定することができる。被測定物内部の各反射面における反射率だけでなく、更に多くの情報が得られることにより光CTで組織診断を行うことが原理的には可能になる。新たに開発した計測系は、広帯域光を用いた干渉計から得られる信号の時間変化を計測することで揺らぎスペクトルを求める。技術的な面で重要なことは、この揺らぎ信号は光計測において不可避な雑音である散射雑音より小さいということである。したがって、従来、だれも光CTにおいて揺らぎスペクトルの計測に成功していなかったが、計測系を二重化して相関を計測する手法を我々が開発し、散射雑音より遥かに小さい信号の計測が可能になった。
我々の研究では、厚さ0.1mmの油膜の両面の熱揺らぎスペクトル、カバーガラスのそれぞれの面に塗った水と油の熱揺らぎスペクトル、私の指内部の熱揺らぎスペクトル、アユの眼球表面近辺の熱揺らぎスペクトルなどを測定し、本計測が原理的に可能であることを示した。
Optical coherence tomography (OCT) provides the depth resolved images of semi-transparent objects non-invasively and is useful especially in medicine. It characterizes each cross section of the onion-like-multi-layer materials by its reflectance. The basic physics principle underlying the OCT is the low-coherence interferometry, which is combined with lateral scanning of the light beam to produce the cross sections of materials. It is clearly desirable to obtain more detailed information regarding each cross section. We have developed a system which measures the fluctuation spectra at all depth in low-coherence interferometry. The atoms and molecules that make up all objects perform random motion with thermal energy. By measuring the spectrum of this thermal motion, physical properties such as viscosity and surface tension of the object can be measured. By providing more information for each cross section, this can be effective in tissue characterizations. The newly developed measurement system determines the fluctuation spectrum by measuring the time change of the signal obtained from the interferometer using broadband light. From the technical point of view, it is important that this fluctuation signal is smaller than the shot-noise, which is the inevitable noise in light measurement. Therefore, conventionally, no one has succeeded in measuring the fluctuation spectrum using the OCT, but we have developed the method to measure the correlation by dualizing the measurement system, and the fluctuation signal much smaller than the shot-noise become detectable.
In our study, we have measured the thermal fluctuation spectrum of both sides of a 0.1-mm-thick oil film, water and oil painted to each side of the cover glass, inside my finger, and the surface of the eyeball of Ayu, and showed the validity of this measurement.
