Abstract 
細胞周期依存的蛍光プローブ(Fucci)導入大腸がん細胞HCT116をNOGマウスに移植し得られた凍結担がん肝組織を用いて、組織切片腫瘍部位における細胞周期別の代謝物質局在について検討を行った。Fucci-green蛍光陽性腫瘍部位をS/G2/M期、Fucci-red蛍光陽性腫瘍部位をG1期として質量イメージング画像と重ね合わせて解析を行った結果、G1期でATP, NADH, UDP-GlcNAc等が顕著に高く解糖系の亢進が示唆された。また、細胞周期同調培養で得られた細胞周期毎の細胞メタボローム解析等により、1)細胞外流出の乳酸がG1期で高く、2)G2/M 期のATP生成はミトコンドリア呼吸依存的であることが認められた。以上から、がん細胞は細胞分裂するたびに内因性機構により酸化ストレスに曝露されること、グルタチオン等の内因正抗酸化機構を阻害することによって増殖を制御出来る可能性が示唆された。
