| Abstract |
２つの課題を中心に研究を進めた。⑴ストレプトアビジン２次元結晶膜（SA 膜）作成法の開発，および⑵ SA 膜表面への結合によるビオチン化タンパク質の安定化を利用した１分子原子間力顕微鏡（AFM）イメージング法の開発である。 課題⑴についてはこれまでの方法をリファインし，かなりの程度隙間無く基板（シリコンウエハ）表面をSA ２次元結晶で覆うことができるようになった。また，AFM走査パラメータの適正化などにより，市販のAFM プローブをそのまま用い，再現性のよいSA 結晶の高分解能イメージングが可能となった。課題⑵では，カタラーゼやフェリチンなどをビオチン化してSA膜２次元結晶面に結合固定し，AFM イメージングを試みた。SA 膜はビオチン化タンパク質固定のための理想的基盤であり，これを利用した１分子AFM イメージングおよびタンパク質を利用した素子開発へと今後の展開が期待される。
